Levulan Kerastick + PDT for Actinic Keratosis
What You Need to Know Before You Apply
What is the purpose of this trial?
This trial tests a new treatment for actinic keratosis (AK), which are rough, scaly patches on the skin caused by sun exposure. The study examines the effectiveness of a reformulated version of Levulan Kerastick (a topical solution) combined with blue light therapy in clearing these skin lesions on the arms. Participants will receive either the active treatment or a placebo. The main goal is to determine if the AK spots disappear completely after 12 weeks. The trial is suitable for individuals in good health with 4-8 mild to moderate AK lesions on one arm, who can maintain a consistent skincare routine and avoid tanning. As a Phase 3 trial, this study represents the final step before FDA approval, offering participants a chance to contribute to the potential availability of a new treatment.
Do I have to stop taking my current medications for the trial?
The trial requires you to stop using certain topical therapies on the treatment area within 2 days to 8 weeks before starting, and some systemic therapies within 6 weeks to 6 months before starting. If you're using phototoxic or photoallergic medications, you need to stop them 8 weeks before starting. It's best to discuss your specific medications with the trial team.
Is there any evidence suggesting that this treatment is likely to be safe for humans?
Previous studies have shown that Levulan Kerastick, used with photodynamic therapy (PDT), offers promising safety results for treating actinic keratosis (AK). Research indicates that patients generally tolerate this treatment well. Most side effects are mild and temporary, with skin pain during the light treatment being the most common issue, though it usually resolves quickly. Other reactions, such as redness or swelling, also tend to be short-lived.
Levulan Kerastick has a strong safety record when used on the face and scalp for AKs. Its FDA approval for these areas supports its safety. The combination of Levulan Kerastick with blue light treatment has proven effective, yielding excellent cosmetic results and side effects that diminish over time. Overall, while the treatment may cause some discomfort, it has consistently been found safe for patients.12345Why do researchers think this study treatment might be promising for actinic keratosis?
Unlike other treatments for actinic keratosis, which often rely on topical creams or cryotherapy, the reformulated Levulan Kerastick with photodynamic therapy (PDT) offers a unique approach by combining a medication with a specialized light treatment. This therapy involves applying Levulan, allowing it to incubate, and then activating it with blue light, which targets and destroys precancerous cells. Researchers are particularly excited because this method could potentially provide more precise and effective treatment with fewer side effects, offering new hope for patients with this common skin condition.
What evidence suggests that this treatment might be an effective treatment for actinic keratosis?
Research has shown that Levulan Kerastick, combined with blue light therapy, effectively treats actinic keratosis (AK), which are rough, scaly patches on the skin. In this trial, participants will receive either the Levulan application or a vehicle application, both followed by blue light therapy. Studies have demonstrated that the Levulan treatment can clear up to 100% of AK spots, completely removing these patches. Patients treated with Levulan and blue light report good cosmetic results, with no scarring or changes in skin color. The FDA has approved this combination, confirming its effectiveness and safety for mild to moderate AK. Overall, the evidence strongly supports its use in effectively clearing AK spots.12367
Are You a Good Fit for This Trial?
This trial is for adults aged 18-85 who have 4-8 mild to moderate actinic keratosis (AK) spots on one arm. Participants must be in good health, avoid sunbathing or tanning, and agree to use effective birth control if needed. They should not use other skin products on the treatment area before visits.Inclusion Criteria
Timeline for a Trial Participant
Screening
Participants are screened for eligibility to participate in the trial
Treatment
Participants receive Levulan application followed by blue light exposure for up to 2 PDT sessions depending on lesion clearance
Follow-up
Participants are monitored for safety, effectiveness, and recurrence of lesions after treatment
What Are the Treatments Tested in This Trial?

Trial Overview
The study tests a new version of Levulan Kerastick (aminolevulinic acid gel) plus blue light therapy versus a placebo gel with blue light. Patients are randomly assigned to either group and may receive up to two treatments depending on how well their AK spots clear.
