What is the purpose of this trial?
This trial explores whether a combination of oral estrogen and progestin, specifically Norethindrone Acetate-Ethinyl Estradiol, can stimulate the release of oxytocin, a hormone linked to bonding and stress relief, in individuals with certain hormone deficiencies. It includes two groups: those with hypopituitary disease, which affects hormone production, and those with a specific hormone deficiency called AVP-D. Participants will take a single dose of the medication, and researchers will check their hormone levels. This trial suits individuals diagnosed with AVP-D or hypopituitary disease who have stable hormone replacement therapy. As an Early Phase 1 trial, this research focuses on understanding how the treatment works in people, offering participants a chance to contribute to groundbreaking medical discoveries.
Will I have to stop taking my current medications?
If you are currently taking estrogen/progestin, you will need to stop for at least 6 weeks before the main study visits. For other medications, there should be no changes within 2 weeks of enrollment or within 5 half-lives of the medication.
Is there any evidence suggesting that this treatment is likely to be safe for humans?
Research has shown that norethindrone acetate and ethinyl estradiol are generally safe for people. These ingredients, commonly found in birth control pills, have been used safely for many years. Other studies have indicated that a single dose of these hormones does not cause significant side effects, and participants did not report major health problems after using them.
However, since this trial is in its early stages, researchers are closely monitoring the treatment's safety. Being in an early phase means limited information exists on its specific effects for oxytocin deficiency. Based on their use in other treatments, these hormones are usually safe. Are You a Good Fit for This Trial?
This trial is for children and teens with certain pituitary disorders, like AVP-D or hypopituitarism, who have stable hormone treatments and can speak English. People can't join if they have a history of blood clots, certain cancers, serious heart problems, recent pregnancy or breastfeeding, severe kidney/liver disease, active smoking, or major mental health issues.Inclusion Criteria

Treatment
Participants receive a single administration of norethindrone acetate and ethinyl estradiol, with blood and saliva sampling at baseline and 24 hours post-administration
Follow-up
Participants are monitored for changes in neurophysin-1 and oxytocin levels, as well as neuropsychological assessments
