What is the purpose of this trial?
This trial explores how psilocybin, a compound found in certain mushrooms, paired with mindfulness therapy, might help people with PTSD, a condition causing severe stress after trauma. Researchers aim to determine if this combination can improve PTSD symptoms by examining changes in brain activity. Participants will receive either psilocybin with standard support or with added mindfulness-based therapy. The trial seeks individuals diagnosed with moderate to severe PTSD who have maintained a stable dose of antidepressants without major changes for a month. As a Phase 2 trial, this research focuses on measuring the treatment's effectiveness in an initial, smaller group of people.
Do I have to stop taking my current medications for the trial?
You may need to stop certain medications before joining the trial. Specifically, you cannot be on monoamine oxidase inhibitors, tricyclic antidepressants, certain non-SSRI or non-SNRI antidepressants, antipsychotics, mood stabilizers, or psychotropic medicines that interact with psilocybin. However, you can continue a stable dose of SSRI antidepressants or sedatives/hypnotics. The study team will review your medications to ensure safety.
Is there any evidence suggesting that this trial's treatments are likely to be safe?
Earlier studies have found psilocybin to be generally safe and well-tolerated when used with psychological support. Research showed that a 25 mg dose did not cause any unexpected side effects, indicating likely safety. Participants also experienced improvements in PTSD symptoms.
Mindfulness-Based Cognitive Therapy (MBCT) is considered a promising method for treating PTSD. Often used alongside other treatments, it has effectively reduced PTSD symptoms.

Are You a Good Fit for This Trial?
This trial is for adults aged 21-65, assigned male or female at birth, with moderate to severe PTSD and depression. Participants must have a BMI between 18-35, weigh at least 60 kg, be on stable antidepressant medication if applicable, and use reliable birth control during and after the study.Inclusion Criteria

Open label administration of 25 mg synthetic Psilocybin combined with standard PAT psychological support plus MBCT in ten participants with PTSD

Trial Overview
The study compares one dose of psilocybin given with either standard psychological support or with mindfulness-based cognitive therapy (MBCT) in people with PTSD. Participants are randomly assigned to each group. Brain changes will also be measured using scans.
