Long-acting Antibodies for Ulcerative Colitis
(SKYLINE-LTE Trial)
What You Need to Know Before You Apply
What is the purpose of this trial?
This trial aims to evaluate the effectiveness of new long-acting antibodies for people with ulcerative colitis (UC). It tests different treatments—SPY001, SPY002, and SPY003—either alone or in combination, to determine which might reduce symptoms. Individuals who participated in a previous UC study and experienced some benefit or require further symptom relief may be suitable candidates for this trial. As a Phase 2 trial, the research focuses on assessing the treatment's effectiveness in an initial, smaller group of participants.
Do I have to stop taking my current medications for the trial?
Yes, you will need to stop taking certain medications, including immunosuppressants, systemic tacrolimus, and any marketed advanced therapy, among others, as they are not allowed during the trial.
Is there any evidence suggesting that this trial's treatments are likely to be safe?
In a previous study, patients with ulcerative colitis tolerated SPY001 well. The treatment's safety matched that of other drugs in its class, which is a positive sign. Another study found no serious side effects related to the treatment. For SPY002, 41.7% of patients experienced side effects, but only 3 were linked to the drug, and just 2 were serious, indicating that most side effects were not severe. Earlier research also showed that patients tolerated SPY003 well, with no reports of serious side effects related to the treatment. Overall, earlier studies have shown these treatments to be manageable in terms of safety.12345
Why are researchers excited about this trial's treatments?
Researchers are excited about SPY001, SPY002, and SPY003 for ulcerative colitis because these treatments use long-acting antibodies, which could mean fewer doses and more sustained symptom relief compared to current treatments like mesalamine, corticosteroids, or biologics. Unlike standard therapies that often target inflammation broadly, these investigational drugs specifically target the immune components causing the inflammation, potentially offering more precise control of the disease. Additionally, the combination regimens being tested could offer even greater benefits by addressing multiple pathways involved in ulcerative colitis, potentially leading to improved outcomes for patients.
What evidence suggests that this trial's treatments could be effective for ulcerative colitis?
Research has shown that SPY001, SPY002, and SPY003 could be promising treatments for ulcerative colitis (UC). In this trial, participants will receive one of these treatments. Specifically, SPY001 reduced a key inflammation score by 9.2 points, outperforming some current treatments. SPY002 reduced this score by 10.7 points and improved colon appearance in 42% of patients. SPY003 helped 40% of patients achieve symptom relief and improved colon appearance in 51% of cases. These results suggest these treatments might be effective options for people with UC.12467

Are You a Good Fit for This Trial?
This trial is for people with ulcerative colitis who took part in a previous study (SPY123-201) and either finished it with benefit, completed certain parts, or needed extra help due to worsening symptoms. People can't join if they have serious infections, use certain immune-suppressing drugs, or have other major health problems.Inclusion Criteria

Trial Overview
The study looks at the long-term safety and effectiveness of three new antibody treatments (SPY001, SPY002, SPY003), used alone or together, in people with ulcerative colitis. It follows up participants from an earlier related trial.
