This trial aims to determine if a single dose of psilocybin, a psychedelic compound, can improve anhedonia (loss of interest or pleasure) in individuals who continue to experience symptoms despite taking antidepressants like SSRIs or SNRIs. Researchers will compare psilocybin's effects to a placebo to assess its impact on brain function and mood. Individuals may qualify if they have major depressive disorder, experience emotional blunting, and have not fully responded to antidepressants. Participants must have no recent experience with psychedelics and must commit to multiple MRI sessions. As a Phase 1 trial, the research focuses on understanding how the treatment works in people, offering participants a chance to be among the first to receive this innovative treatment.

Do I have to stop taking my current medications for the trial?

The trial does not specify that you must stop taking your current prescription medications, but you must refrain from taking all non-prescription medications and supplements for at least 1 week before the psilocybin session unless approved by the Investigator.

This trial is for adults who still experience lack of pleasure or emotional numbness (anhedonia) even though they are already taking SSRI or SNRI antidepressants. Participants should not have conditions that would make psilocybin unsafe, but specific exclusions aren't listed here.

Inclusion Criteria

Have an identified support person
Able to provide voluntary signed and dated informed consent
State willingness to comply and be available for all study requirements, including psychological, cognitive, imaging and procedural evaluations for the duration of the study

Exclusion Criteria

Any other physical or psychological symptom, medication, or relevant finding making participant unsuitable based on clinical judgment
Contraindications to MRI scanning (implantable devices, bone hardware, various IUD)
Current or previous psychiatric conditions meeting DSM-5 criteria for psychotic disorders (schizophrenia, schizoaffective disorder, MDD with psychosis), bipolar 1 or 2 disorder, or current diagnosis of active substance use disorder
