RVX-sCPD9 Booster for COVID-19
What You Need to Know Before You Apply
What is the purpose of this trial?
This trial tests a new COVID-19 booster called RVX-sCPD9, administered as a nasal spray. Researchers aim to assess its safety and the body's response at various dose levels. The trial is open to healthy adults who have completed a full COVID-19 vaccine series and received at least one booster, with the last dose given at least 16 weeks prior. Participants should not have had a recent COVID-19 infection or certain health conditions, such as asthma or heart disease. As a Phase 1 trial, participants will be among the first to receive this new treatment, aiding researchers in understanding its effects in people.
Do I have to stop taking my current medications for the trial?
The trial does not specify that you must stop taking your current medications. However, you cannot use intranasal medications, including steroids, for 30 days before and after the study product administration, and for 6 months after for intranasal steroids. It's best to discuss your specific medications with the trial team.
Is there any evidence suggesting that RVX-sCPD9 is likely to be safe for humans?
Earlier studies have shown that the RVX-sCPD9 vaccine, administered as a nasal spray, is safe. Research indicates that the most common side effect was a runny nose, occurring in about 3.7% of the 15,500 recipients. About 8% of participants reported other mild reactions, such as nausea. These studies found no serious safety issues, suggesting the vaccine is well-tolerated. This offers some confidence in its safety for potential trial participants.12345
Why are researchers excited about this trial's treatment?
Researchers are excited about the RVX-sCPD9 vaccine for COVID-19 because it is delivered intranasally, offering a needle-free alternative that could increase vaccine accessibility and compliance. Unlike traditional COVID-19 vaccines, which are typically administered via injection, this vaccine uses a nasal atomization device to deliver the dose directly to the nasal passages, potentially enhancing the immune response where the virus often enters the body. Additionally, the varying dosages tested (ranging from 1x10^2 to 5x10^4 FFU) allow researchers to determine the most effective dose for generating a strong immune response, which could lead to more efficient vaccination strategies.
What evidence suggests that RVX-sCPD9 could be an effective booster for COVID-19?
Research has shown that RVX-sCPD9, a COVID-19 vaccine administered nasally, is promising in generating strong immune responses. Studies have found that RVX-sCPD9 can build robust immunity with just one dose, outperforming some existing COVID-19 vaccines. This trial will test different doses of the RVX-sCPD9 vaccine, with participants receiving varying amounts to assess immune response. The vaccine is designed to activate both antibody and cell-based immune responses, which are crucial for fighting the virus. Early results indicated that participants who received higher doses experienced fewer symptomatic COVID-19 cases. These initial findings suggest that RVX-sCPD9 could effectively boost immunity against COVID-19.13467
Are You a Good Fit for This Trial?
This trial is for healthy adults aged 18-64 who have already received a COVID-19 vaccine. Participants must not be pregnant, must use effective birth control if of childbearing potential, and should have stable health with no recent major changes in chronic conditions.Inclusion Criteria

Trial Overview
The study tests the safety and immune response of four different dose levels of an intranasal COVID-19 booster vaccine called RVX-sCPD9. All participants receive one dose through the nose; there is no placebo group.

Experimental Treatment
Healthy adults will receive a single dose of 5x10\^4 FFU RVX-sCPD9 vaccine. It will be administered intranasally through a nasal atomization device with a spray divider to provide 0.25mL in each nostril. 3 sentinel participants, under 50 years of age, will be enrolled first and then the reminder. N= 20
