What You Need to Know Before You Apply

What is the purpose of this trial?

This trial tests a new gene therapy treatment called RP-A701 for individuals with dilated cardiomyopathy linked to the BAG3 gene. The goal is to assess the safety and effectiveness of this treatment when administered as a single intravenous dose. It targets adults diagnosed with this heart condition who experience frequent symptoms despite following treatment guidelines. Participants should have had an ICD (implantable cardioverter-defibrillator) for at least three months and stable heart failure symptoms. As a Phase 1 trial, this research aims to understand how the treatment works in people, offering participants the opportunity to be among the first to receive this innovative therapy.

Do I have to stop taking my current medications for the trial?

The trial does not specify if you need to stop taking your current medications. However, you must have been on a stable heart failure treatment plan for at least 30 days before joining the trial.

Is there any evidence suggesting that RP-A701 is likely to be safe for humans?

Research has shown that RP-A701 is a new gene therapy being tested for BAG3-associated dilated cardiomyopathy (BAG3-DCM). This early-stage study primarily aims to determine the treatment's safety for people. Since RP-A701 is being tested in humans for the first time, no prior information exists on human reactions.

The study is in its initial phase, focusing on safety testing. Early trials like this are designed to closely monitor participants for any side effects. The trial's commencement indicates it has already passed initial safety checks in the lab and animal studies, which is encouraging. However, potential side effects in humans are still under investigation. Participants will be closely monitored to ensure the treatment's safety.12345

Why do researchers think this study treatment might be promising?

Unlike the standard treatments for dilated cardiomyopathy, which often include medications like beta-blockers and ACE inhibitors, RP-A701 offers a novel gene therapy approach. This treatment works by delivering a specific gene directly into the heart cells, aiming to correct the underlying genetic cause of the disease rather than just manage the symptoms. Researchers are excited about RP-A701 because it has the potential to provide a long-lasting solution by targeting the root cause, offering hope for more effective and sustained improvement in heart function.

What evidence suggests that RP-A701 might be an effective treatment for dilated cardiomyopathy?

Research has shown that RP-A701 is a promising gene therapy for treating BAG3-related dilated cardiomyopathy (DCM), a heart condition caused by a gene mutation. In this trial, participants will receive a single intravenous dose of RP-A701. This experimental treatment aims to correct the genetic issue causing the disease. Although limited data exists on its effectiveness in humans, gene therapies like RP-A701 are designed to deliver correct copies of faulty genes to improve heart function. Early studies on similar gene therapies have shown promise in addressing genetic problems in heart diseases. While still in the early stages of testing, this treatment offers hope for tackling the root cause of this heart condition.23467

Are You a Good Fit for This Trial?

This trial is for adults aged 18–65 with a genetic form of dilated cardiomyopathy (BAG3 variant), mild to moderate heart dysfunction, and stable heart failure symptoms. Participants must not have severe artery blockages, uncontrolled high blood pressure, or other major heart conditions.

Trial Overview

The study tests RP-A701, a gene therapy delivered by IV that uses a harmless virus to carry the BAG3 gene into the body. The main goal is to see if it's safe and tolerable in people with BAG3-related dilated cardiomyopathy.
