GB-4362 + Enfortumab Vedotin + Pembrolizumab for Bladder Cancer
What You Need to Know Before You Apply
What is the purpose of this trial?
This trial tests a new drug, GB-4362, combined with enfortumab vedotin and pembrolizumab, to determine its safety and tolerability for individuals with advanced bladder cancer. GB-4362 aims to reduce the side effects of enfortumab vedotin, a chemotherapy drug. The study seeks participants already scheduled to receive enfortumab vedotin and pembrolizumab for their bladder cancer treatment. Participants should not have had previous treatment with certain drugs or have uncontrolled diabetes. As a Phase 1 trial, this research focuses on understanding how the treatment works in people, offering participants the opportunity to be among the first to receive this new drug combination.
Do I have to stop taking my current medications for the trial?
The trial protocol does not specify if you need to stop taking your current medications. However, you cannot have received anti-cancer treatment with chemotherapy, biologics, or investigational agents within 4 weeks before starting the trial.
Is there any evidence suggesting that this trial's treatments are likely to be safe?
Research has shown that combining enfortumab vedotin and pembrolizumab is generally safe. Various studies have found this combination effective and well-tolerated, with most side effects being manageable through treatment or adjustments.
GB-4362, a newer drug, aims to reduce some side effects of enfortumab vedotin, such as nerve damage that can cause pain or numbness. Since GB-4362 is in the early testing phase (Phase 1), information on its safety remains limited. Early trials primarily focus on understanding safety, and more information will emerge as the study progresses.
Overall, enfortumab vedotin with pembrolizumab has been safely used in many patients, while researchers continue to carefully study GB-4362 for safety when used with these drugs.12345Why are researchers excited about this trial's treatments?
Researchers are excited about the combination of GB-4362, Enfortumab Vedotin, and Pembrolizumab for bladder cancer because this treatment approach targets cancer cells in a unique way. Unlike most treatments that focus solely on one mechanism, this combination therapy uses Enfortumab Vedotin to deliver a powerful chemotherapy agent directly to cancer cells, while Pembrolizumab, an immune checkpoint inhibitor, helps the immune system recognize and attack the cancer. GB-4362 is a novel component that may enhance the overall effectiveness of the treatment by working synergistically with the other drugs. This multi-pronged approach has the potential to improve outcomes for patients who have limited options with current standard treatments.
What evidence suggests that this trial's treatments could be effective for bladder cancer?
Studies have shown that combining enfortumab vedotin and pembrolizumab effectively treats advanced bladder cancer. This combination has been linked to longer survival rates and manageable side effects. Research indicates that patients with varying kidney function levels experience similar positive results. In this trial, participants will receive enfortumab vedotin, pembrolizumab, and the new drug GB-4362. GB-4362 is designed to reduce side effects from enfortumab vedotin, such as nerve damage. Although limited human data exists on GB-4362, early findings suggest it might help by neutralizing harmful parts of the treatment. Overall, the current combination of enfortumab vedotin and pembrolizumab has shown promise, and adding GB-4362 aims to enhance safety.12456
Are You a Good Fit for This Trial?
This trial is for adults (18+) with advanced or metastatic urothelial cancer who weigh at least 50 kg, have a life expectancy of at least 3 months, and are healthy enough to participate. People can't join if they've had certain prior treatments, uncontrolled diabetes, active brain tumors, severe allergies to monoclonal antibodies, or serious ongoing side effects.Inclusion Criteria
