Indomethacin for Metabolic Syndrome
What You Need to Know Before You Apply
What is the purpose of this trial?
This trial aims to examine how the drug Indomethacin affects brain blood flow in individuals with Metabolic Syndrome, a condition characterized by high blood pressure, high blood sugar, excess body fat around the waist, and abnormal cholesterol levels. Researchers seek to determine if these effects differ between males and females. Participants will either have Metabolic Syndrome or serve as healthy controls, receiving both Indomethacin and a placebo at different times. Individuals with at least three symptoms of Metabolic Syndrome, such as high waist size and high blood sugar, may qualify for the trial. As an Early Phase 1 trial, this research focuses on understanding how Indomethacin works in people, offering participants an opportunity to contribute to groundbreaking medical knowledge.
Do I need to stop taking my current medications for the trial?
The trial requires that you do not take medications known to influence cardiovascular function, except for contraceptive hormones. This includes medications for blood pressure, diabetes, and cholesterol.
Is there any evidence suggesting that Indomethacin is likely to be safe for humans?
Research has shown that the FDA has approved indomethacin for treating conditions like acute pain and arthritis, indicating its safety for these uses. However, some risks exist. Indomethacin may increase the risk of blood clots, heart attacks, or strokes, particularly with long-term use. It can also cause stomach and digestive issues and affect kidney function. These side effects occur more frequently with extended use, so short-term use in a trial might pose less risk. Always weigh the benefits and risks before deciding to join a study.12345
Why do researchers think this study treatment might be promising for Metabolic Syndrome?
Indomethacin is unique because, unlike other treatments for Metabolic Syndrome that typically focus on managing individual symptoms like high blood pressure, glucose levels, or cholesterol, it targets inflammation, which is thought to play a central role in the syndrome. This anti-inflammatory action represents a potential new approach to addressing the root causes of Metabolic Syndrome. Researchers are excited about indomethacin's potential to provide a more comprehensive treatment option by possibly reducing the overall inflammation associated with the condition, potentially improving multiple symptoms at once.
What evidence suggests that Indomethacin might be an effective treatment for Metabolic Syndrome?
Research has shown that indomethacin reduces inflammation and pain in conditions like rheumatoid arthritis by lowering swelling and fever. Although primarily used for these conditions, no direct evidence yet supports indomethacin's effectiveness for metabolic syndrome or its symptoms. This trial will explore indomethacin's potential effects on metabolic syndrome, including its influence on blood flow and inflammation. Participants will receive either a placebo followed by indomethacin or indomethacin followed by a placebo. Results from studies on various conditions have been mixed, indicating the need for further research to understand its impact on metabolic syndrome.12567

Are You a Good Fit for This Trial?
This trial is for adult men and women, both healthy and those with metabolic syndrome. Participants must not have high blood pressure or obesity, should have normal blood sugar and cholesterol levels (unless they meet criteria for metabolic syndrome), and women need regular menstrual cycles. Oral contraceptives are allowed.Inclusion Criteria

Trial Overview
The study compares how a drug called indomethacin versus a placebo affects brain blood flow in people with or without metabolic syndrome, using MRI scans. It also looks at differences between males and females.
How Is the Trial Designed?

Treatment groups
Experimental Treatment
Male and female participants, age-matched with Healthy Controls, who have at least 3 defining symptoms of Metabolic Syndrome. Randomized to either Placebo then Indomethacin or Indomethacin then Placebo for 2 MRI visits.
Male and female participants 18-45 without Metabolic Syndrome. On study for 3 visits (up to 6 months). Randomized to either Placebo then Indomethacin or Indomethacin then Placebo for 2 MRI visits.
