What You Need to Know Before You Apply

What is the purpose of this trial?

This trial tests two drugs, Avutometinib and Defactinib, to determine if they can enhance treatment for glioblastoma, a type of brain cancer. The trial consists of two stages: the first administers the drugs before standard radiation therapy, while the second combines them with radiation for certain patients. The primary goal is to assess whether the drugs can be safely administered before and during radiation. Candidates for this trial likely have a high-grade brain tumor and plan to receive radiation therapy as part of their treatment. As an Early Phase 1 trial, this research aims to understand how the treatment works in people, offering participants the opportunity to be among the first to receive these potentially groundbreaking drugs.

Is there any evidence suggesting that this trial's treatments are likely to be safe?

Research has shown that the combination of avutometinib and defactinib is generally safe. One study on this treatment found no major safety issues for participants. Patients with various types of cancer, including brain tumors, have tolerated it well.

Additionally, this combination has been tested in other conditions and received fast-track FDA approval for a specific type of ovarian cancer, indicating relative safety. However, as this trial is in the early stages, the treatment's safety continues to be carefully monitored and evaluated.12345

Why are researchers excited about this trial's treatments?

Researchers are excited about Avutometinib and Defactinib for glioblastoma because these drugs target cancer cells in a novel way. Unlike traditional treatments that mainly focus on surgery, radiation, and chemotherapy, Avutometinib and Defactinib work by inhibiting specific proteins involved in cancer cell survival and growth. This mechanism is different from the standard approaches and offers a new strategy to tackle aggressive tumors. Additionally, the oral administration of these drugs could offer a more convenient option compared to intravenous therapies.

What evidence suggests that Avutometinib and Defactinib could be effective for glioblastoma?

Research shows that the drugs avutometinib and defactinib can reach glioblastoma tumors and affect their growth. In this trial, participants will receive these drugs in different stages. Other studies have shown promise in treating similar conditions, with up to 31% of patients responding in some types of cancer. This suggests they might help slow tumor growth or even shrink tumors. The drugs target specific parts of cancer cells, potentially stopping their growth. Although data on their use in glioblastoma remains limited, these early findings offer hope for effective treatment.13567

Are You a Good Fit for This Trial?

This trial is for adults with newly diagnosed high-grade gliomas, including glioblastoma. Participants must have not started radiation yet and need to be healthy enough for study drugs and standard treatment. Only those with a specific tumor type (MGMT-unmethylated GBM) continue in the second stage.

Exclusion Criteria

I have used acid-reducing medications in the last 2 days.
I have had brain or scalp cancer treated with radiation before.
I have had a blood clot or stroke in the past 90 days.

What Are the Treatments Tested in This Trial?

Interventions

  • Avutometinib
  • Defactinib
  • Radiation Therapy

Trial Overview

The study tests two oral drugs, avutometinib and defactinib, given before and during standard brain radiation therapy. The first part gives one cycle of these drugs before radiation; the second part continues them alongside radiation for certain patients.
